It originated from shooting three-pointers in basketball and wishing for it to go in or chanted as a celebration after scoring a three-pointer. Very similar to just shouting "three" or "get in". It can also be used in contexts where desirable things are in sight and the achievement of aforementioned things must be celebrated.
Similar hype words include lfg, we take those.
Hand gestures indicating you have just shot a three-pointer must be accompanied by saying this phrase.

"Hey John, these presale opportunities are available for a bargain!"
"Wow, you're right. Give me three (gesture as if just shot a three-pointer)"

I don't give three fucks

When the person in question, does not have the capacity to actually care about what you are saying, but does have bit of intrest. So the amount of fucks given cannot be great.

So they might give two.

Person 1: Do you wanna know about this REALLY funny thing?!

Person 2: I DON'T GIVE THREE FUCKS

a phrase that nobody uses, but is a very good way to show how you do not care, two shits is an understatement.

A term originated in Boston demonstrating disdain for any activity, person, or thing. Often used in conjunction as a hand wave with three fingers. In contrast to the positive take saying three's up.
